EP-1233 is an experimental multi-epitope DNA vaccine designed for the prevention of HIV-1 infection. Developed by Epimmune (later acquired by Pharmexa), the vaccine utilizes a plasmid DNA vector to deliver a "polytope" sequence encoding 21 distinct cytotoxic T-lymphocyte (CTL) epitopes derived from six HIV-1 proteins: Gag, Pol, Env, Vpr, Rev, and Nef. These epitopes were selected for their conservation across HIV-1 clades and their ability to bind to common HLA class I molecules, aiming to elicit a broad and durable cellular immune response. EP-1233 was primarily evaluated in Phase I clinical trials, such as HVTN 053, often as part of a heterologous prime-boost strategy involving recombinant viral vectors like MVA-mBN32. Development of the candidate ceased following the acquisition of Epimmune by Pharmexa and the lack of progression beyond early-stage trials.
