Overview

EP-302 is a preclinical-stage targeted membrane-disrupting peptide (tMDP) developed by Esperance Pharmaceuticals (and later assigned to A28 Therapeutics) for the treatment of various cancers. It consists of a nucleolin-binding moiety conjugated to CLIP-71, a novel cationic alpha-helical lytic peptide. EP-302 selectively targets and binds to nucleolin, which is overexpressed on the surface of cancer cells and tumor endothelial cells. Upon binding, the lytic peptide domain disrupts the cell membrane, inducing rapid cell death primarily via necrosis. Preclinical studies have demonstrated its efficacy in breast and prostate cancer xenograft models.
