Overview

EP547 is a potent, highly selective small molecule antagonist of the Mas-related G protein-coupled receptor X4 (MRGPRX4). It is being developed as an oral, once-daily therapy for the treatment of cholestatic and uremic pruritus—debilitating itch conditions associated with chronic liver diseases such as primary biliary cholangitis (PBC) and primary sclerosing cholangitis (PSC), as well as chronic kidney disease. MRGPRX4 is expressed on sensory itch neurons in the skin and can be activated by bile acids, bilirubin, and related metabolites that accumulate in these diseases. By blocking MRGPRX4 activation, EP547 aims to provide targeted relief from pruritus without affecting the underlying liver or kidney disease. Clinical studies have shown that EP547 is safe and well-tolerated[1][2][6][8].
