Overview

A combination regimen including **epacadostat**, an oral, selective indoleamine 2,3-dioxygenase 1 (IDO1) inhibitor; **nivolumab**, a PD-1 immune checkpoint inhibitor; and **chemotherapy**. This regimen is being investigated primarily for advanced solid tumors, including squamous cell carcinoma of the head and neck (SCCHN), non-small cell lung cancer (NSCLC), melanoma, colorectal cancer, ovarian cancer, glioblastoma, and diffuse large B-cell lymphoma (DLBCL). Epacadostat works by inhibiting IDO1, an enzyme that suppresses antitumor immunity in the tumor microenvironment, while nivolumab blocks the PD-1 receptor on T cells, releasing PD-1 pathway–mediated inhibition of anti-tumor immune responses. Chemotherapy provides a standard cytotoxic effect and may enhance immunogenicity of cancer cells[1][2][5][7].
