EPC-003 is an investigational, fully human, bispecific, armored CAR-T cell therapy targeting B7-H3 (CD276) and interleukin-13 receptor subunit alpha-2 (IL13Rα2), being developed by Elpis Biopharmaceuticals for the treatment of glioblastoma and glioblastoma multiforme.[3][5][7][9][11][12][14][15] It incorporates dual antigen targeting against CD276 and IL13Rα2 to enhance tumor specificity and reduce target escape, and is further engineered with a secreted multi-functional “armor” designed to boost CAR-T cell persistence, overcome immunosuppression in the tumor microenvironment, and enable penetration of the blood–brain barrier.[1][3][7][9] In preclinical orthotopic glioblastoma models, EPC-003 induced rapid tumor regression after intracranial administration and showed activity following intravenous dosing, and it is now being evaluated in an investigator-initiated phase 1 trial delivering repeated intraventricular doses via an Ommaya reservoir in patients with recurrent or refractory glioblastoma.[1][5][7][12][13]
