EpCAM antibodies are a class of monoclonal antibodies and antibody-derived constructs designed to target the Epithelial Cell Adhesion Molecule (EpCAM), also known as CD326. EpCAM is a 40 kDa transmembrane glycoprotein that is highly overexpressed in a wide variety of carcinomas, including colorectal, gastric, breast, and lung cancers, while showing restricted expression in normal epithelial tissues. This differential expression makes it an attractive target for both therapeutic and diagnostic applications. Therapeutically, EpCAM antibodies function through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), or as bispecific T-cell engagers (BiTEs) that bridge tumor cells and T-cells to induce lysis. Historically, edrecolomab was one of the first monoclonal antibodies approved for cancer treatment, and catumaxomab was approved for the treatment of malignant ascites. In addition to therapy, EpCAM antibodies are the gold standard for the enrichment and detection of circulating tumor cells (CTCs) in liquid biopsy platforms.
