Overview

EpCAM CAR T-cells are chimeric antigen receptor (CAR) T-cell therapies engineered to target Epithelial Cell Adhesion Molecule (EpCAM), a cell-surface glycoprotein highly expressed in various epithelial cancers, including gastric, colorectal, ovarian, and breast cancers. These autologous or allogeneic T-cells are genetically modified to express a CAR consisting of an anti-EpCAM single-chain variable fragment (scFv) linked to intracellular signaling domains (such as CD3ζ and costimulatory domains like CD28, 4-1BB, or Dectin-1). Upon binding to EpCAM on tumor cells, the CAR T-cells are activated to proliferate, secrete cytokines, and exert direct cytotoxic activity against the target cancer cells. EpCAM CAR T-cells are being evaluated in preclinical and early-phase clinical trials for the treatment of EpCAM-positive solid tumors.
