Overview

This is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting the epithelial cell adhesion molecule (EpCAM), developed by Sichuan University at West China Hospital. The therapy utilizes a second-generation CAR construct incorporating a Dectin-1 costimulatory domain, delivered via lentiviral transduction under GMP conditions. EpCAM is a transmembrane glycoprotein highly expressed in various epithelial malignancies, including nasopharyngeal, breast, and gastric cancers, while having limited expression in normal tissues. The treatment is being evaluated for safety and efficacy in patients with advanced EpCAM-positive solid tumors, with administration routes including intravenous infusion and intraperitoneal injection for cases with peritoneal metastasis. Clinical results published in *Science Advances* (2023) demonstrated a manageable safety profile and evidence of anti-tumor activity, including partial responses and prolonged progression-free survival in some patients with refractory solid tumors.
