Drug intelligence / Profile preview
EpCAM-directed CAR T cells are an experimental, preclinical cell therapy consisting of T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting epithelial cell adhesion molecule (EpCAM), a tumor-associated antigen highly expressed on epithelial-derived tumors and metastases. Developed by researchers at Ludwig-Maximilians-University Munich (LMU Munich), this adoptive T-cell immunotherapy has been investigated in preclinical models of lung cancer brain metastases. In syngeneic mouse models, local (intraparenchymal) administration of these CAR T cells demonstrated significant intratumoral accumulation, reduced tumor growth, and prolonged survival, whereas systemic (intravenous) administration showed limited efficacy. To overcome the immunosuppressive tumor microenvironment and enhance CAR T-cell persistence, researchers have explored combination strategies, including CSF1R inhibition to deplete tumor-associated macrophages and microglia (TAM/M) or co-administration with anti-angiogenic agents (anti-VEGF-A/VEGF-R2).
