Overview

This combination regimen includes **epcoritamab** (a subcutaneous bispecific CD3xCD20 antibody), **polatuzumab vedotin** (an antibody-drug conjugate targeting CD79b), **rituximab** (a CD20 monoclonal antibody), **cyclophosphamide** (an alkylating agent), **doxorubicin** (an anthracycline cytotoxic agent), and **prednisone** (a corticosteroid). This approach is under clinical investigation as first-line treatment for newly diagnosed diffuse large B-cell lymphoma (DLBCL). Epcoritamab enhances T cell-mediated cytotoxicity by binding CD3 on T cells and CD20 on B cells; polatuzumab vedotin delivers a microtubule-disrupting cytotoxin to CD79b+ B cells; rituximab mediates B cell lysis through anti-CD20 targeting; cyclophosphamide and doxorubicin introduce cytotoxic damage to cancer cells; prednisone suppresses inflammation and immune responses. The combination aims to further improve response rates and depth of remission over conventional immunochemotherapy.
