Epetraborole is a novel, investigational small molecule antibiotic that inhibits leucyl-tRNA synthetase (LeuRS), an essential enzyme responsible for attaching leucine to its corresponding tRNA, thereby blocking protein synthesis in bacteria. It has demonstrated potent activity against a range of Gram-negative bacteria and nontuberculous mycobacteria, including *Mycobacterium abscessus* and *Burkholderia pseudomallei*. Epetraborole has been evaluated in clinical trials for the treatment of bacterial infections such as urinary tract infections, intestinal infections, community-acquired infections, and melioidosis. The drug exhibits favorable pharmacokinetic properties including low plasma protein binding, extensive tissue distribution, and linear kinetics following both intravenous and oral administration[1][5][6][7][8].
