Overview

EPGN696 is an orally available small-molecule antagonist of lysophosphatidic acid receptor 1 (LPA1), originally developed by Epigen Biosciences and subsequently licensed to Novo Nordisk for further development in kidney and other fibrotic/metabolic diseases.[3][8][15][16] By selectively blocking LPA1 signaling, EPGN696 is designed to inhibit lysophosphatidic acid–driven pathways involved in fibrosis, inflammation, and growth factor responses, and has shown preclinical efficacy and safety in rodent models of kidney disease and other fibrotic settings, including preservation of proximal tubule structure and function and reduction of tubulointerstitial fibrosis.[1][6][9][11] The compound is part of a broader LPA1 antagonist program targeting conditions such as diabetic nephropathy, idiopathic pulmonary fibrosis, and nonalcoholic steatohepatitis, with current development status reported as preclinical and no human clinical trials initiated to date.[2][3][11]
