Overview

This biologic therapy involves genetically modifying autologous T cells to express a CAR that recognizes EphA2, a receptor tyrosine kinase overexpressed in various solid tumors, including glioblastoma, non-small cell lung cancer (NSCLC), and pediatric bone tumors like osteosarcoma. The CAR construct typically includes an EphA2-specific single-chain variable fragment (scFv), costimulatory domains (such as CD28 or 4-1BB), and a CD3ζ signaling domain. Clinical and preclinical development is being pursued by institutions such as Xuanwu Hospital and Memorial Sloan Kettering Cancer Center, with early-phase trials investigating its safety and efficacy in recurrent glioblastoma.
