Overview

EphA3-CAR-T is an autologous chimeric antigen receptor T-cell immunotherapy in preclinical development that targets ephrin type-A receptor 3, a tumor-associated antigen expressed on glioblastoma, diffuse midline glioma, and other high-grade glioma cells. The therapy uses genetically engineered T cells expressing an anti-EphA3 single-chain variable fragment derived from anti-EphA3 monoclonal antibodies such as 3C3-1 or IIIA4 with a second-generation CAR architecture incorporating CD28 costimulation to drive T-cell activation and cytotoxic killing of EphA3-positive tumor cells. Published preclinical work indicates activity against intracranial glioma models and suggests the engineered T cells can traffic to brain tumors, but no commercial brand name or late-stage clinical development program has been clearly identified; the program appears to be associated primarily with academic development including Mayo Foundation for Medical Education and Research.
