Overview

EphA3 CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the Ephrin type-A receptor 3 (EphA3), a protein frequently overexpressed in glioblastoma (GBM) and associated with brain cancer stem cells and tumor vasculature. Developed by researchers at institutions including QIMR Berghofer and the Garvan Institute, these CAR T cells have demonstrated curative efficacy in orthotopic xenograft models of glioblastoma. They function by infiltrating the tumor, disaggregating tumor cells, and inducing apoptosis in patient-derived neurospheres and explant organoids. The therapy aims to address the poor prognosis of GBM by targeting chemo-resistant tumor-propagating cells and the heterogeneous tumor microenvironment.
