Overview

EPHB4-CAR-T cells are an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target the Ephrin type-B receptor 4 (EPHB4), a member of the ephrin receptor subfamily of tyrosine kinases. Developed by the National Cancer Center Hospital in Japan, these CAR-T cells are produced using a non-viral gene transfer system, which is intended to offer a safer and potentially more cost-effective alternative to traditional viral vector-based methods. EPHB4 is frequently overexpressed in various malignancies, including Ewing sarcoma and other advanced solid tumors, where it contributes to tumor cell survival, proliferation, and angiogenesis. Upon binding to EPHB4 on the tumor cell surface, the CAR-T cells undergo activation and expansion, leading to the direct cytotoxic destruction of the cancer cells. The therapy is currently being investigated in Phase I clinical trials (such as the CARTiEr trial) to assess its safety, kinetics, and preliminary efficacy in patients with relapsed or refractory EPHB4-positive solid tumors.
