Overview

EphB4 siRNA-DOPC is a preclinical therapeutic candidate consisting of a small interfering RNA (siRNA) targeting erythropoietin-producing hepatocellular receptor B4 (EphB4) encapsulated in neutral 1,2-dioleoyl-sn-glycero-3-phosphatidylcholine (DOPC) liposomes. Developed primarily by researchers at the University of Texas MD Anderson Cancer Center and the University of Puerto Rico, this agent is designed to silence EphB4 expression in tumor cells. EphB4 is a receptor tyrosine kinase overexpressed in various cancers, including ovarian and breast cancers, where it promotes tumor cell survival, migration, invasion, and angiogenesis, and acts as an alternative receptor for erythropoietin (Epo) to mediate Epo-induced tumor growth. Preclinical studies in orthotopic mouse models of ovarian and breast cancer have demonstrated that EphB4 siRNA-DOPC effectively downregulates EphB4 expression, induces tumor cell apoptosis, and inhibits tumor growth, particularly when combined with chemotherapy such as docetaxel.
