ephrin A5-Fc is a recombinant fusion protein consisting of the extracellular domain of the ephrin-A5 ligand fused to the Fc region of human IgG. It functions as a high-affinity ligand for the Eph family of receptor tyrosine kinases, with a particular focus on EphA3, which is frequently overexpressed in glioblastoma (GBM). By binding to and activating EphA3, ephrin A5-Fc triggers the ERK/MAPK signaling pathway, which induces the differentiation of cancer stem cells and reduces tumor cell proliferation. In preclinical research, it has demonstrated the ability to attenuate tumor growth and prevent the formation of invasive intracranial xenografts. While primarily utilized as a research tool to study cell adhesion, migration, and axon guidance, its potential to modulate oncogenic signaling in aggressive brain tumors has made it a subject of therapeutic investigation.
