EphrinA1.Fc1 is a recombinant fusion protein consisting of the EphA2 receptor ligand, EphrinA1, genetically fused to the constant domain (Fc) of human IgG1. Developed at Wake Forest University School of Medicine, this biologic is designed to target the EphA2 receptor, a membrane-associated biomarker that is overexpressed in the majority of Glioblastoma Multiforme (GBM) cases but absent in normal brain tissue. The Fc fusion enhances the stability and binding affinity of the EphrinA1 ligand. Preclinical studies have demonstrated that EphrinA1.Fc1 can successfully bind to GBM specimens and activate the EphA2 receptor, eliciting cellular responses such as cell rounding. It has been investigated as a targeting moiety for the delivery of therapeutics and as a diagnostic probe when radiolabeled with isotopes like Iodine-125. Research suggests that using EphrinA1.Fc1 in combination with other GBM-restricted ligands, such as those targeting IL13Rα2, may help overcome tumor heterogeneity and improve therapeutic coverage in GBM patients.
