Epi-039 is a cationic lipid nanoparticle composed of 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP) and folate, developed by EpicentRx in collaboration with the University of California San Diego (UCSD). It is designed to encapsulate adenoviral vectors, such as the oncolytic adenovirus AdAPT-001, to facilitate cellular entry into tumor cells that lack or have low expression of the coxsackievirus and adenovirus receptor (CAR). By targeting folate receptors, which are overexpressed on various cancer cells, Epi-039 enables CAR-independent transduction of tumors. Additionally, the liposomal encapsulation shields the viral vector from neutralizing antibodies in the bloodstream, potentially allowing for effective systemic administration of adenoviral gene therapies.
