EPI-12323

EPI-12323 (brand name: Naturasone) is a small molecule, non-glucocorticoid steroid developed by Epigenesis Pharmaceuticals as a once-daily inhaled treatment for asthma and chronic obstructive pulmonary disease (COPD). Its mechanism of action is classified as adenosine deaminase (ADA) inhibition, which targets the inflammatory and airway obstruction cascade in the irritated or asthmatic lung. The drug has demonstrated pronounced anti-inflammatory and bronchodilatory activity in preclinical models. It reached Phase IIA clinical trials, including a randomized, double-blind, parallel-group study evaluating its efficacy versus placebo in symptomatic moderate-to-severe asthma patients maintained on low-dose inhaled corticosteroids. Licensed from academia as a new molecular entity, it was intended as a potential replacement for conventional inhaled corticosteroids, but development appears to have been discontinued.
