Overview

**epidermal growth factor receptor small interfering RNA** is a synthetic double-stranded RNA specifically designed to target and silence the expression of the epidermal growth factor receptor (EGFR) gene using the mechanism of RNA interference (RNAi). By binding to EGFR mRNA, the small interfering RNA (siRNA) mediates its degradation, leading to reduced EGFR protein expression. EGFR is a cell surface receptor with intrinsic tyrosine kinase activity, involved in cell proliferation, survival, and signaling (PI3K, MAPK, PKC pathways), frequently dysregulated in various cancers. Downregulation of EGFR via siRNA has demonstrated inhibitory effects on tumor cell proliferation, migration, and enhanced sensitivity to chemotherapy and radiotherapy in preclinical models, particularly in non-small cell lung cancer, esophageal squamous cell carcinoma, ovarian cancer, and glioma[1][2][3][6]. The development of effective delivery systems, such as lentiviral vectors and functionalized nanogels, has been a significant focus to enable targeted and efficient in vivo application[1][6][7].
