Drug intelligence / Profile preview
epigallocatechin gallate + copper + bacillus Calmette-Guérin (EGCG-Cu@BCG) is a novel synergistic metal-phenolic nanomaterial complex developed for the treatment of bladder cancer, particularly non-muscle-invasive bladder cancer (NMIBC). It is synthesized by coating the surface of Bacillus Calmette-Guérin (BCG) with an epigallocatechin gallate-copper (EGCG-Cu) coordination complex via a gentle physical incubation method. EGCG-Cu@BCG is designed to overcome the limitations of BCG monotherapy, such as limited direct tumor-killing ability and variable immune activation, by combining the direct cytotoxic effects of EGCG-Cu with the immunoadjuvant properties of BCG. The complex induces tumor cell apoptosis, mitochondrial membrane potential decrease, and strong immunogenic cell death (ICD), characterized by calreticulin (CRT) exposure on the cell membrane and high mobility group box 1 (HMGB1) release from the nucleus. In preclinical mouse models of bladder cancer, intravesical administration of EGCG-Cu@BCG has demonstrated enhanced anti-tumor efficacy and reduced tumor recurrence without significant systemic toxicity.
