Epirubicin + oxaliplatin + capecitabine is a three-drug chemotherapy combination regimen commonly referred to as EOX (or sometimes EXE). It consists of: - Epirubicin, an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and RNA synthesis. - Oxaliplatin, a platinum-based chemotherapeutic agent that forms crosslinks in DNA, inhibiting DNA replication and transcription. - Capecitabine, an oral prodrug converted to 5-fluorouracil (5-FU) in the body; it inhibits thymidylate synthase and interferes with DNA synthesis. This combination is primarily used as first-line therapy for advanced or metastatic gastric cancer and gastroesophageal junction adenocarcinoma. Clinical trials have shown that EOX provides comparable or superior efficacy compared to older regimens such as epirubicin/cisplatin/5-FU (ECF), with the added convenience of oral administration for capecitabine[1][2][4]. The regimen is typically administered every three weeks for up to eight cycles[1][2].
