Overview

This is a combination regimen consisting of three drugs—epirubicin, pertuzumab, and trastuzumab—used primarily in the neoadjuvant (pre-surgical) or adjuvant (post-surgical) treatment of HER2-positive breast cancer. Epirubicin is an anthracycline chemotherapy agent that intercalates DNA and inhibits topoisomerase II, leading to cytotoxicity. Pertuzumab and trastuzumab are monoclonal antibodies targeting the human epidermal growth factor receptor 2 (HER2); they bind to different domains on HER2, blocking receptor dimerization and signaling pathways that promote tumor cell proliferation. The dual blockade with pertuzumab and trastuzumab has shown synergistic antitumor activity when combined with chemotherapy such as epirubicin[5][6]. This regimen is used for patients with HER2-overexpressing early-stage or locally advanced breast cancer as part of a multi-agent protocol.
