EpiTAC-ADC is a dual-modality therapeutic platform developed by EpiBiologics that combines extracellular targeted protein degradation (eTPD) with antibody-drug conjugate (ADC) technology. The platform utilizes bispecific antibodies where one arm targets a specific pathogenic cell-surface protein, such as the receptor tyrosine kinase c-MET, and the other arm binds to a tissue-enriched degrading receptor. This binding facilitates the selective internalization and degradation of the target protein through endosomal-lysosomal or proteosomal pathways. Simultaneously, the EpiTAC-ADC delivers a cytotoxic payload to the cell, providing a secondary mechanism of action. This dual approach is intended to overcome limitations of current therapies, such as acquired resistance or dependence on high target expression, and has shown efficacy in preclinical models across a range of c-MET expression levels, including those with low and medium expression that do not rely exclusively on oncogenic signaling.
