Overview

EPL-1410 is a novel, orally active small molecule dual inhibitor of Indoleamine 2,3-dioxygenase 1 (IDO1) and Tryptophan 2,3-dioxygenase (TDO2), developed by Emcure Pharmaceuticals. It is designed for use in immuno-oncology to counteract the immunosuppressive environment created by tumors through tryptophan catabolism. By inhibiting both IDO1 and TDO2, EPL-1410 prevents the depletion of tryptophan and the excessive generation of kynurenine pathway metabolites, which otherwise suppress T-cell activity. In preclinical studies, EPL-1410 demonstrated significant dose-dependent efficacy in reducing tumor volume in mouse syngeneic models of colon carcinoma (CT-26) and melanoma (B16F10), showing high oral bioavailability and a favorable safety profile.
