EpMab-37-mG2a-f is a defucosylated mouse IgG2a monoclonal antibody targeting the epithelial cell adhesion molecule (EpCAM), a transmembrane glycoprotein frequently overexpressed in various carcinomas and associated with poor prognosis. Developed using the Cell-Based Immunization and Screening (CBIS) method, this antibody is a modified version of the original EpMab-37 (mouse IgG1, kappa). The defucosylation of the Fc region is designed to enhance its binding affinity to Fcγ receptors on immune effector cells, thereby significantly increasing antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have demonstrated that EpMab-37-mG2a-f exhibits potent ADCC and complement-dependent cytotoxicity (CDC) against EpCAM-expressing breast and pancreatic cancer cell lines. Furthermore, administration of the antibody has shown significant suppression of xenograft tumor development in vivo, suggesting its potential as a therapeutic regimen for EpCAM-positive malignancies.
