Epopromycin B is a small molecule proteasome inhibitor derived from natural products, specifically isolated from *Streptomyces* species. Developed by Hunan Dino Pharmaceutical, it is structurally characterized by an epoxyketone functional group, which allows it to form a covalent bond with the N-terminal threonine residues of the catalytic subunits within the 20S proteasome. By inhibiting the proteasome's activity, Epopromycin B disrupts protein degradation pathways, leading to the accumulation of misfolded proteins and the induction of apoptosis in malignant cells. It has been investigated in Phase 1 clinical trials in China for the treatment of advanced solid tumors to assess its safety, pharmacokinetics, and preliminary antitumor activity.
