Overview

Epothilone D (BMS-241027) is a small-molecule microtubule stabilizer developed for the treatment of Alzheimer's disease and previously investigated in oncology. Its mechanism of action involves binding to and stabilizing microtubules, counteracting the destabilization caused by hyperphosphorylated tau protein seen in tauopathies such as Alzheimer's disease. By stabilizing microtubules, Epothilone D aims to preserve neuronal structure and function, potentially reducing cognitive decline. Preclinical studies demonstrated that BMS-241027 could reverse behavioral deficits and reduce neuropathology in mouse models of tauopathy. The drug is brain-penetrant and was considered safer than other compounds in its class for CNS indications. Clinical development for Alzheimer's disease reached Phase 1 but was discontinued; oncology development was also halted due to safety concerns compared with newer analogs[1][2][5][6].
