EPPS (4-(2-hydroxyethyl)-1-piperazinepropanesulfonic acid) is a small molecule drug candidate being developed by Korea Pharma for the treatment of dementia, specifically Alzheimer's disease. Originally known as a common laboratory buffer (HEPPS), research conducted by the Institute for Basic Science (IBS) in South Korea demonstrated that EPPS can directly bind to and disaggregate amyloid-beta (Aβ) plaques in the brain. Unlike many therapeutic candidates that aim to prevent the formation of Aβ aggregates, EPPS is designed to break down existing plaques into monomers, potentially reversing neurotoxicity. In preclinical animal models, oral administration of EPPS led to the clearance of Aβ aggregates and the recovery of cognitive function. Korea Pharma acquired the technology for EPPS and is currently advancing it through preclinical development.
