Epratuzumab is a humanized monoclonal antibody that targets CD22, a cell surface glycoprotein expressed on mature B-cells and many malignant B-cells. It binds with high specificity to the third Ig-like domain of CD22, modulating B-cell function by perturbing the B-cell receptor (BCR) signaling complex and intensifying the inhibitory role of CD22. This leads to reduced signaling and diminished activation of B cells, resulting in partial depletion of circulating B-cells while minimizing impact on overall immune function. Epratuzumab acts primarily as an immunomodulatory agent rather than inducing acute cytotoxicity. It was initially developed for non-Hodgkin lymphoma and leukemia but has also been investigated for systemic lupus erythematosus (SLE) and Sjögren’s disease[2][3][5][6][8].
