Overview

**Epratuzumab + rituximab** is a combination of two monoclonal antibodies targeting distinct B-cell antigens, CD22 and CD20, respectively. Epratuzumab is a humanized anti-CD22 antibody (hLL2, LymphoCide), acting as an immunomodulator by modulating B-cell activity without causing extensive B-cell depletion. Rituximab is a chimeric anti-CD20 antibody (Rituxan) that induces cytotoxic effects on B cells via complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and apoptosis, leading to lysis of CD20-expressing B lymphocytes. The combination has been explored in clinical trials for relapsed/refractory non-Hodgkin’s lymphoma (NHL), including follicular lymphoma and diffuse large B-cell lymphoma, where it has shown a favorable safety profile and promising clinical activity[1][3][4][5][7]. The rationale is complementary mechanisms: epratuzumab’s immunomodulation with rituximab’s cytotoxicity may enhance antitumor efficacy while maintaining tolerability[2][4].
