epratuzumab + 90Y-epratuzumab is a radioimmunotherapy (RIT) regimen consisting of the humanized monoclonal antibody epratuzumab and its yttrium-90-labeled counterpart. Developed by Nantes University Hospital in collaboration with Immunomedics (now part of Gilead), this combination targets CD22, a cell surface antigen highly expressed on B-cells. The regimen typically involves sequential injections where unlabeled epratuzumab is administered to saturate non-tumor binding sites or modulate the target, followed by 90Y-epratuzumab, which delivers targeted beta-radiation to malignant B-cells. This localized radiation induces cell death primarily through ionizing radiation-induced DNA damage. The therapy has been evaluated in Phase 1/2 clinical trials for the treatment of relapsed or refractory CD22-positive B-cell acute lymphoblastic leukemia (B-ALL).
