EPS-3903 is a novel, potent, and selective oral small molecule inhibitor of Signal Transducer and Activator of Transcription 6 (STAT6), currently in preclinical development by Enanta Pharmaceuticals. STAT6 is a key transcription factor in the IL-4 and IL-13 signaling pathways, which are central to type 2 immune-driven inflammatory diseases such as atopic dermatitis. Preclinical data have demonstrated that EPS-3903 exhibits nanomolar binding potency and high selectivity, achieving over 90% inhibition of phosphorylated STAT6 (pSTAT6) in vivo following oral administration. Designed for once-daily dosing, EPS-3903 is being developed as a potential oral alternative to injectable biologics like dupilumab. Enanta Pharmaceuticals is currently conducting IND-enabling activities and targets an Investigational New Drug (IND) filing in the second half of 2026.
