Epsin siRNA nanoparticle (S2PNP-siEpsin1/2) is a preclinical RNA interference (RNAi) therapy designed to treat atherosclerosis. It consists of small interfering RNAs (siRNAs) targeting the endocytic adaptor proteins Epsin 1 and Epsin 2, encapsulated within nanoparticles functionalized with a lesional macrophage-specific targeting peptide (S2P). In atherosclerotic plaques, Epsins 1 and 2 facilitate CD36-mediated lipid uptake and the degradation of ABCG1, leading to foam cell formation and inflammation. By silencing these targets, the nanoparticle therapy promotes plaque regression, reduces pro-inflammatory cytokine expression, and ameliorates dyslipidemia. Research also suggests that targeting liver Epsins can inhibit PCSK9-mediated degradation of the low-density lipoprotein receptor (LDLR), further improving cholesterol clearance.
