Epsin siRNA nanoparticles (also referred to as S2PNP-siEpsin1/2) are an experimental nanomedicine platform designed to deliver small interfering RNAs (siRNAs) targeting Epsin 1 and Epsin 2. Epsins are endocytic adaptor proteins that, in the context of atherosclerosis, facilitate CD36-mediated lipid uptake and promote the degradation of the cholesterol transporter ABCG1 in macrophages. In the liver, Epsins contribute to PCSK9-mediated degradation of the low-density lipoprotein receptor (LDLR). By silencing Epsin 1 and Epsin 2 via RNA interference, these nanoparticles aim to reduce inflammation, enhance cholesterol efflux, and lower circulating LDL-C levels, thereby halting or reversing atherosclerotic plaque progression. The platform often utilizes a stabilin-2 targeting peptide (S2P) for macrophage-specific delivery or nanoparticle encapsulation for liver-directed therapy.
