Drug intelligence / Profile preview
The Epstein-Barr virus (EBV) gp350 ferritin nanoparticle vaccine is an investigational prophylactic vaccine designed to prevent EBV infection and its associated complications, such as infectious mononucleosis, multiple sclerosis, and various malignancies (e.g., Hodgkin lymphoma, Burkitt lymphoma, and nasopharyngeal carcinoma). Developed by the National Institute of Allergy and Infectious Diseases (NIAID) Laboratory of Infectious Diseases in collaboration with the NIAID Vaccine Research Center, the vaccine consists of the EBV envelope glycoprotein gp350 fused to self-assembling Helicobacter pylori non-heme ferritin. This fusion protein self-assembles into a spherical nanoparticle that displays the gp350 antigen in a dense, highly immunogenic array. The vaccine is formulated with Novavax's saponin-based Matrix-M (or Matrix-M1) adjuvant to enhance immune responses. In Phase I/II clinical trials, the vaccine has demonstrated a favorable safety profile and elicited robust neutralizing antibody titers and polyfunctional CD4+ and CD8+ T-cell responses in both EBV-seropositive and EBV-seronegative healthy adults.
