The Epstein-Barr virus gp350-ferritin vaccine is a nanoparticle-based investigational vaccine designed to prevent infection by the Epstein-Barr virus (EBV) and its associated clinical manifestations, such as infectious mononucleosis. Developed by the NIAID Vaccine Research Center (VRC), the vaccine utilizes a self-assembling ferritin nanoparticle platform to display the EBV envelope glycoprotein gp350 in a multivalent fashion. gp350 is the most abundant protein on the surface of the virus and is the primary ligand used by EBV to attach to and infect B lymphocytes via the CD21 receptor. By presenting gp350 on a nanoparticle, the vaccine mimics the repetitive surface structure of a virus, which significantly enhances the induction of potent neutralizing antibodies compared to monomeric protein vaccines. It is currently being evaluated in early-phase clinical trials to determine its safety, tolerability, and immunogenicity in healthy adults.
