Epstein-Barr virus-specific stem cell memory cytotoxic T lymphocytes (EBV-specific T_SCM-enriched T cells) represent an advanced form of adoptive T-cell therapy. This therapy involves the ex vivo expansion of a patient's or donor's T cells, specifically enriching for those with a stem cell memory (T_SCM) phenotype that are specific to Epstein-Barr virus (EBV) antigens. T_SCM cells are characterized by superior self-renewal capacity, robust engraftment, prolonged persistence, and enhanced anti-cancer efficacy compared to more differentiated T-cell subsets. The primary mechanism of action is to restore or augment the immune system's ability to recognize and eliminate EBV-infected cells, which are often implicated in various malignancies and post-transplant lymphoproliferative disorders (PTLD). This approach aims to provide long-lasting immunity and therapeutic effect against EBV-associated diseases.
