Eptazocine is a benzomorphan derivative opioid analgesic developed by Nihon Iyakuhin Kogyo (now known as Nichi-Iko Pharmaceutical) in Japan. It was first synthesized in 1976 and approved for marketing in Japan in 1987. Eptazocine acts as a mixed κ-opioid receptor agonist and μ-opioid receptor antagonist, providing effective analgesia with fewer side effects typical of μ-opioid agonists such as respiratory depression and addiction potential. It is primarily used for moderate to severe pain management including cancer pain and postoperative pain. The drug is commonly administered as its hydrobromide salt via oral or injectable routes. Common adverse effects include nausea, vomiting, sweating, fever, thirst, injection site reactions, dizziness, and sedation. Due to its opioid nature it carries risk of dependence.
