Equator Therapeutics is developing a first-in-class small molecule thermogenic program aimed at treating obesity and metabolic disorders. The drug candidates are designed to increase energy expenditure by modulating mitochondrial thermogenesis, specifically targeting the ADP/ATP carrier (AAC) and Uncoupling Protein 1 (UCP1). By promoting a controlled proton leak across the inner mitochondrial membrane, these compounds uncouple mitochondrial respiration from ATP synthesis, thereby dissipating energy as heat. This mechanism mimics the natural thermogenic function of brown adipose tissue and is intended to provide a systemic approach to weight loss and improved metabolic health by increasing the basal metabolic rate.
