Equecabtagene autoleucel is a fully human, autologous chimeric antigen receptor T-cell (CAR-T) therapy that targets B-cell maturation antigen (BCMA). It is developed using lentiviral vectors to transduce the patient’s own T cells with a CAR construct containing a fully human single-chain variable fragment (scFv), CD8a hinge and transmembrane domains, and 4-1BB co-stimulatory plus CD3ζ activation domains. The therapy is indicated primarily for relapsed or refractory multiple myeloma and has demonstrated high efficacy and durable responses in heavily pretreated patients. Equecabtagene autoleucel has also received orphan drug status for multiple myeloma and is being investigated for autoimmune diseases such as myasthenia gravis and neuromyelitis optica[1][3][4][6].
