ERβ mRNA-LNP is a preclinical mRNA-based therapy consisting of estrogen receptor β (ERβ) mRNA encapsulated in lipid nanoparticles (LNPs). Developed by researchers at the Houston Methodist Research Institute, it is designed to induce tumor-specific upregulation of ERβ, a receptor with known anti-tumor and anti-metastatic properties that is typically downregulated or absent in aggressive breast cancers. In preclinical models of inflammatory breast cancer (IBC) and triple-negative breast cancer (TNBC), direct administration of ERβ mRNA-LNPs resulted in high tumor levels of the receptor, inhibition of metastasis, slowed tumor growth, and a less immunosuppressive tumor microenvironment characterized by reduced M2-like macrophages and increased cytotoxic T cell activity.
