Overview

ErA-TM is an engineered variant of the bacterial L-asparaginase enzyme derived from *Erwinia chrysanthemi*, designed to retain strong L-asparaginase activity while exhibiting greatly reduced L-glutaminase co-activity. This modification aims to decrease the significant non-immune toxicities associated with standard L-asparaginase therapies for acute lymphoblastic leukemia (ALL). ErA-TM is created via specific site-directed mutations in the enzyme's active site, resulting in a safer therapeutic profile with reduced side effects such as weight loss and glutaminase-mediated toxicities, while maintaining antileukemic efficacy comparable to existing asparaginase formulations. Recent modifications (e.g., fusion with albumin binding domain) have aimed to improve its in vivo stability and persistence. Its primary indication is as an alternative to standard L-asparaginase for the treatment of ALL, especially in settings where toxicity is a major limiting factor.[1][4]
