ERAS-12 is an EGFR D2/D3 bispecific monoclonal antibody under discovery and preclinical development by Erasca, intended to inhibit wildtype EGFR signaling mediated by overexpression of EGFR, which may play a role in cancer therapy pathways[1][9]. By binding two distinct domains of EGFR, ERAS-12 is designed to potentially address escape routes in RAS/MAPK pathway signaling, which are implicated in the development of resistance to existing therapies in RAS/MAPK pathway-driven cancers.
