ERAS-2 is a preclinical drug candidate developed by Erasca, designed as a potent and selective inhibitor of the Human Epidermal Growth Factor Receptor 2 (HER2). As part of Erasca's MAPKlamp strategy, which aims to comprehensively shut down the RAS/MAPK pathway, ERAS-2 was intended to target HER2-driven cancers, including those with HER2 amplifications or mutations. HER2 is a well-validated oncogenic driver that activates downstream signaling through the MAPK and PI3K pathways. By inhibiting HER2, ERAS-2 aims to block these proliferative signals and induce apoptosis in tumor cells. While initially listed in Erasca's discovery-stage pipeline, the program has seen less public development in recent years as the company prioritized other candidates like ERAS-007 (ERK inhibitor) and ERAS-601 (SHP2 inhibitor).
