Overview

ERAS-4 is a small molecule drug developed as a pan-KRAS inhibitor, specifically targeting the KRAS G12D mutation. It was initially developed by Erasca as part of their internal RAS/MAPK pathway-focused pipeline. The mechanism of action involves inhibiting the activity of mutant KRAS proteins, which are key drivers in various solid tumors such as colorectal cancer, non-small cell lung cancer, and pancreatic ductal adenocarcinoma. The program aimed to address resistance and tolerability challenges seen with other RAS pathway inhibitors by directly targeting oncogenic KRAS alterations. However, development of ERAS-4 has been discontinued; some molecules from this program may serve as backup compounds for newer candidates like ERAS-4001 under licensing agreements[1][2][4].
