ErbB3 siRNA is a research-stage RNA interference (RNAi) modality designed to selectively silence the expression of the ErbB3 (HER3) receptor. ErbB3 is a member of the epidermal growth factor receptor (EGFR) family that, while lacking intrinsic kinase activity, plays a critical role in oncogenic signaling by heterodimerizing with other family members like ErbB2 (HER2) or EGFR. By targeting ErbB3 mRNA for degradation via the RNA-induced silencing complex (RISC), this siRNA inhibits the translation of the receptor, thereby disrupting downstream PI3K/AKT signaling pathways essential for tumor cell survival, proliferation, and motility. It has been investigated in various preclinical models, including lung adenocarcinoma, cutaneous melanoma, and cervical cancer, demonstrating significant tumor growth inhibition in xenograft mouse models.
