Overview

Eremophil-1(10),11(13)-dien-12,8α-olide is a sesquiterpene lactone of the eremophilane class, primarily isolated from plants such as *Inula helenium* (Elecampane) and various *Ligularia* species. It is characterized by an α-methylene-γ-lactone moiety, which is the key pharmacophore responsible for its biological activity. This moiety acts as a Michael acceptor, forming covalent bonds with the thiol groups of cysteine residues in target proteins. Research has demonstrated that this compound possesses significant anti-inflammatory and cytotoxic properties, largely mediated through the inhibition of the NF-κB signaling pathway and the induction of apoptosis in cancer cells. While it is not an approved pharmaceutical drug, it is a constituent of herbal extracts used in traditional medicine and dietary supplements for respiratory and inflammatory conditions.
